Terminally differentiated neurons are non-proliferating, long-lived cells with high metabolic activity. As a result, they must deal with a considerable DNA damage burden. The way neurons cope with DNA damage differs strikingly from mitotic or cycling cells. Recent studies highlight how DNA damage and dysfunctional DNA repair may contribute to neurodegenerative diseases. Yet it is not clear whether genome instability is a primary driver of neuronal loss or an epiphenomenon.
